چکیده
Objectives: To determine the utility of the cell transfer technique (CTT) for the BRAF molecular testing on thyroid fine-needle aspirates (FNA). Methods: Polymerase chain reaction (PCR)based BRAF molecular testing was performed on tissues obtained through CTT from both airdried and ethanol-fixed direct smears of thyroid FNA, and then compared with the corresponding thyroidectomy formalin-fixed, paraffin-embedded (FFPE) tissues on 30 cases. Results: BRAF testing was successfully performed on 29/30 air-dried CTT, 27/30 ethanol-fixed CTT, and 27/30 FFPE. The results exhibited 11, 13, and 13 BRAF mutations and 18, 14, and 14 wild types for the air-dried CTT, the ethanol-fixed CTT, and the FFPE, respectively. The concordance rate was 96% between air-dried and ethanol-fixed CTT, 88% between air-dried CTT and FFPE, and 92% between ethanol-fixed CTT and FFPE. Conclusions: PCR-based BRAF mutational testing can be reliably performed on the direct smears of the thyroid FNA through the application of the CTT.

متن کاملSimultaneously Detection of 50 Mutations at 20 Sites in the BRAF and RAS Genes by Multiplexed Single-Nucleotide Primer Extension Assay Using Fine-Needle Aspirates of Thyroid Nodules.
Fine-needle aspiration (FNA) is commonly used for primary evaluation of thyroid nodules. Twenty to 30 percent of thyroid nodules remain indeterminate after FNA evaluation. Studies show the BRAF p.V600E to be highly specific for papillary thyroid carcinoma (PTC), while RAS mutations carry up to 88 percent positive predictive value for malignancy. We developed a two-tube multiplexed PCR assay fol...
متن کاملAnalysis of BRAF point mutation and RET/PTC rearrangement refines the fine-needle aspiration diagnosis of papillary thyroid carcinoma.
Point mutations in BRAF are genetic hallmarks of papillary thyroid carcinoma (PTC). In this retrospective study, we examined thyroid aspirates and corresponding paraffin-embedded surgical samples for the presence of BRAF mutations. Altogether, we examined 96 cases, including 69 PTCs, 19 follicular adenomas, and eight nontoxic nodular goiters for BRAF; 60 of these samples were also examined for ...
متن کاملProper Indication of BRAFV600E Mutation Testing in Fine-Needle Aspirates of Thyroid Nodules
BACKGROUND The aim of this study was to evaluate the proper indication of adjunctive BRAF(V600E) mutation analysis at the time of ultrasound-guided fine-needle aspiration in the diagnosis of thyroid nodules. METHODS This study included 518 nodules in 479 patients who underwent ultrasound-guided fine-needle aspiration with BRAF(V600E) mutation. We calculated and compared the diagnostic perform...
متن کاملEvaluation of the Anyplex BRAF V600E Real-Time Detection Assay Using Dual-Priming Oligonucleotide Technology in Fine-Needle Aspirates of Thyroid Nodules
BACKGROUND Several molecular assays have been developed to detect the BRAF V600E mutation in fine needle aspirates (FNAs) for the diagnosis of papillary thyroid cancer. Using a multiplex PCR technique, we evaluated the Anyplex BRAF V600E Real-time Detection (Anyplex) assay and compared its efficacy with that of the Seeplex BRAF V600E ACE Detection (Seeplex) method. METHODS We tested 258 conse...
